Both include live Akkermansia muciniphila along with four organisms involved in microbial fermentation, cross-feeding, and short-chain fatty acid biology. Akkermansia has become one of the more closely watched organisms in metabolic microbiome research because of its relationship with the intestinal mucus layer and a growing body of human intervention research.
The five-organism ecosystem also has an interesting clinical history. Pendulum previously studied a closely related formulation in adults with type 2 diabetes and reported improvements in post-meal glucose control. That study supports the biological strategy behind the organism combination, but it was not a direct trial of either today's retail Metabolic Daily or today's Metabolic Daily PRO.
The practical distinction is straightforward: the retail formula offers the same five-organism strategy at 300 million AFU, while PRO provides a higher 1-billion-AFU dose. There is no head-to-head trial showing that PRO produces proportionally greater clinical benefit.
For years, probiotic marketing revolved around a remarkably simple idea: more bacteria must be better.
A bottle containing 50 billion organisms sounded more impressive than one containing 10 billion. Fifteen species looked more sophisticated than five. The numbers became part of the sales pitch even though they told us relatively little about what those organisms were actually doing once they reached the gut.
Microbiome research has made that way of comparing probiotics increasingly outdated.
Different organisms occupy different ecological niches. Some interact with the mucus layer. Some ferment carbohydrates. Some produce short-chain fatty acids. Others consume metabolites produced by neighboring organisms and turn them into something new.
Pendulum's Metabolic Daily platform is built around that more functional view of the microbiome.

This distinction is worth making at the beginning because the names are nearly identical.

PRO therefore provides a little more than three times the total labeled AFU per capsule. That is a formulation difference, not proof of three times the effect. The two products have not been compared head-to-head in a clinical outcomes trial, and probiotic biology does not scale in a simple linear relationship with AFU count.

Why Akkermansia Gets So Much Attention
Akkermansia muciniphila naturally lives close to the intestinal mucus layer, placing it at an interesting interface between the gut microbiome and the epithelial surface beneath it.
Over the past decade, lower Akkermansia abundance has repeatedly been associated with obesity, insulin resistance, type 2 diabetes, and other metabolically unfavorable states. Those observations did not prove that low Akkermansia caused those conditions, but they were consistent enough to justify controlled human intervention studies.
That progression is important. Microbiome research contains thousands of associations. Far fewer organisms have moved into randomized human trials.
The Human Akkermansia Research Is Becoming More Interesting
An early human trial published in Nature Medicine in 2019 studied adults who were overweight or obese and had insulin resistance. Participants received live Akkermansia, pasteurized Akkermansia, or placebo for three months.
The study was small, but the pasteurized preparation produced changes in several metabolic measures, including insulin sensitivity and fasting insulin. More importantly, it demonstrated that Akkermansia could be administered to humans and produce measurable physiologic effects.
In 2026, another randomized study examined pasteurized Akkermansia during weight-loss maintenance. Participants first completed an eight-week weight-loss phase and were then followed for another 24 weeks. Those receiving Akkermansia regained less weight than the placebo group.
A separate 2026 trial in adults with metabolic syndrome was more nuanced. Akkermansia did not significantly improve the study's primary measure of insulin sensitivity across the entire group, but exploratory analyses suggested that people starting with relatively low Akkermansia abundance may have responded differently.
That may eventually become an important part of microbiome medicine. The effect of adding an organism may depend partly on what is already living in the gut.
These studies are highly relevant to the broader Akkermansia story, but they should not be treated as direct trials of either Pendulum Metabolic Daily or Metabolic Daily PRO. Different studies have used different strains, doses, live or pasteurized organisms, and different populations.
The Other Four Organisms Are Part of the Point
Neither Metabolic Daily formula is simply an Akkermansia supplement with four organisms added to fill out the label.
The formulation is designed around microbial interaction.
Clostridium butyricum
Clostridium butyricum is known for producing butyrate, one of the major short-chain fatty acids generated through intestinal fermentation. Butyrate serves as an important energy source for colon cells and participates in normal intestinal-barrier and immune physiology.*
Anaerobutyricum hallii
Anaerobutyricum hallii participates in microbial cross-feeding. It can use metabolites generated by other bacteria and convert them into additional fermentation products, including short-chain fatty acids.
Clostridium beijerinckii
Clostridium beijerinckii is another anaerobic fermenter involved in microbial metabolism. Its presence reflects the effort to assemble organisms with complementary functions rather than relying on one probiotic mechanism.*
Bifidobacterium infantis
Bifidobacterium infantis metabolizes carbohydrates and produces compounds that neighboring organisms can use. Within a microbial ecosystem, that can be just as important as what an organism produces by itself.
Microbial Cross-Feeding: The Part Most Probiotic Labels Never Explain
The idea is simple. One bacterium consumes a substrate and produces a metabolite. Another bacterium uses that metabolite and produces something else.
These exchanges happen throughout the colon. They influence which organisms flourish, which metabolites accumulate, and how the surrounding intestinal environment behaves.
That is closer to the way a microbiome actually functions than the usual image of a capsule depositing a collection of independent bacteria into the gut.
The five-organism Metabolic Daily ecosystem was built around those relationships.
Why Butyrate Matters
Butyrate has become one of the best-known microbial metabolites in gut research.
Colon cells use it as an important fuel source. It also participates in signaling associated with intestinal barrier integrity, immune regulation, and metabolic physiology. Short-chain fatty acids can interact with receptors on intestinal and enteroendocrine cells, creating one of the communication pathways between gut microbes and the rest of the body.*
Pendulum's earlier diabetes research adds an interesting human piece to this biology. A follow-up metabolomic analysis found increased circulating butyrate during treatment with its five-organism research formulation. Changes were also observed in ursodeoxycholic acid, or UDCA.
That does not tell us that every person taking Metabolic Daily will raise circulating butyrate. It does show that a related bacterial consortium was capable of changing measurable microbial metabolites in humans.
The Pendulum Diabetes Trial
Pendulum's best-known clinical study was a multicenter, randomized, double-blind, placebo-controlled trial involving 76 adults with type 2 diabetes, published in 2020.
The experimental formulation was known as WBF-011 and contained the same five bacterial species used across the Metabolic Daily ecosystem:
- Akkermansia muciniphila
- Clostridium butyricum
- Clostridium beijerinckii
- Anaerobutyricum hallii
- Bifidobacterium infantis
Participants receiving WBF-011 had improved post-meal glucose exposure compared with placebo. The investigators also reported an approximately 0.6 percentage point improvement in A1C as a secondary outcome.
For microbiome research, the study was useful because it showed that a deliberately assembled bacterial consortium could produce measurable metabolic effects in humans rather than simply changing the composition of a stool sample.
WBF-011 was not today's retail Metabolic Daily and it was not today's Metabolic Daily PRO. The research formulation used a different dosing strategy and included inulin. The work is more directly connected with Pendulum's Glucose Control product development.
For that reason, neither Metabolic Daily nor Metabolic Daily PRO should be advertised as clinically proven to lower A1C by 0.6%.
The study is better understood as evidence supporting the broader five-species biological strategy.
What Does the Microbiome Have to Do With GLP-1?
GLP-1 is produced naturally by enteroendocrine cells in the intestine. Nutrients, bile acids, short-chain fatty acids, and other microbial metabolites can influence signaling in those cells.
That gives the intestinal microbiome a legitimate role in the physiology surrounding endogenous GLP-1 production.
It should not be confused with the pharmacology of semaglutide, tirzepatide, or other GLP-1-based medications. Those medications produce sustained receptor stimulation at pharmacologic exposures. Microbial signaling works on a different scale.
The microbiome-GLP-1 connection remains worth studying because it is part of normal human metabolic physiology, not because probiotics are natural substitutes for prescription drugs.

What About Weight Management?
The microbiome participates in weight regulation, but body weight is influenced by a much larger network involving appetite, food intake, muscle mass, physical activity, sleep, medications, hormones, genetics, and the food environment.
The recent Akkermansia weight-maintenance research is encouraging because it moves the field further into controlled human studies. The intervention in that trial, however, was a different pasteurized Akkermansia preparation rather than either Pendulum Metabolic Daily product.
Neither Metabolic Daily nor Metabolic Daily PRO should therefore be presented as a stand-alone weight-loss treatment.
The more reasonable role is as part of a broader microbiome and metabolic-health strategy.*
Does the Higher PRO Dose Matter?
Metabolic Daily PRO provides 1 billion AFU compared with 300 million AFU in retail Metabolic Daily. That is a meaningful formulation difference, but we do not yet have the evidence needed to translate that difference into a precise clinical advantage.
Probiotic effects depend on more than the number of viable organisms swallowed. Strain characteristics, survival through the gastrointestinal tract, baseline microbiome composition, diet, substrate availability, dosing consistency, and the ecological environment of the colon can all influence what happens next.
For someone who specifically wants Pendulum's higher-potency professional formulation, PRO provides that option. For someone who wants the same five-organism approach at the standard retail dose, Metabolic Daily provides it in both 30- and 90-capsule sizes.
What we cannot responsibly say is that 1 billion AFU produces 3.3 times the effect of 300 million AFU.
Where the Two Metabolic Daily Formulas Fit
Retail Metabolic Daily is the everyday 300-million-AFU formulation and is available in 30- and 90-capsule sizes.
Metabolic Daily PRO provides the same five organisms at 1 billion AFU per capsule in a 30-capsule professional formulation.
Glucose Control PRO has the closest relationship to Pendulum's published clinical development work involving adults with type 2 diabetes.
GLP-1 Probiotic PRO is positioned more specifically around microbial pathways associated with endogenous GLP-1 signaling.
These formulas overlap enough that taking several at once is not automatically more sophisticated. The useful question is which formulation best matches the reason for using it.

Where We Land
The Metabolic Daily concept is interesting because it moves probiotic formulation away from a simple contest over the biggest organism count.
Live Akkermansia is difficult to manufacture and deliver commercially. Butyrate-producing organisms are biologically relevant. Microbial cross-feeding reflects the way bacteria actually behave in a living ecosystem. A closely related five-species combination has produced measurable metabolic effects in randomized human research, while Akkermansia itself now has an expanding human intervention literature.
There is still plenty we do not know. We do not yet know who benefits most from these specific organisms, whether baseline Akkermansia abundance predicts response in routine clinical use, how long supplementation needs to continue, or whether the higher PRO potency translates into better long-term outcomes.
That is a reasonable place for the science to be. Microbiome medicine is finally moving beyond broad associations and into better human trials. Products like Metabolic Daily are interesting because their design increasingly reflects that science, not because every metabolic question has already been answered.
